1107 TRPV4 activation inhibits inflammatory responses in human macrophages

Aggravation of atopic dermatitis (AD) is closely associated with excessive production cytokines by macrophages. To investigate the mechanisms how macrophages in human skin sense extracellular environment will help to understand diseases, however details remain unclear. In this study, we searched expression pattern 27 transient receptor potential (TRP) channels immune cells using protein atlas data base, and found that TRPV4, a non-selective cation channel high Ca2+ permeability, specifically expressed on monocytes. Pharmacological activation TRPV4 inhibits NFκB signaling, resulting suppression IL-1β primary monocytes derived from Furthermore, inhibited differentiation into GM-CSF (M1) macrophages, but not M-CSF (M2) AD dermis, number iNOS-positive/TRPV4-negative were significantly increased compared healthy samples. These findings highlight physiological relevance for macrophage regulation homeostasis raise possibility may be an anti-inflammatory target.
